In the past two years, incremental advances in standard proteome technology have increased the speed of protein identification with higher levels of automation and sensitivity. Proteins in FFPE material are stable over decades but their efficient extraction and streamlined analysis by mass spectrometry (MS)-based proteomics has so far proven challenging. Herein we describe a MS-based proteomic workflow for quantitative profiling of large FFPE tissue cohorts directly from histopathology glass slides. Mass Spectrometry in Proteomics MS has become a powerful tool in proteomics research to precisely determine the molecular mass of peptides and proteins and the sequences. In tandem mass spectrometry, fragmentation of peptides and proteins gives sequence information for protein identification as well as identification and localization of post-translational or other covalent modifications.
